Helpful Tips for Acclimating Alocasia Tissue Culture Plants

Acclimating Alocasia tissue culture (TC) plants is similar to bringing a baby home from the NICU. These plants have spent their entire lives in a sterile lab environment with 100% humidity and constant access to sugar. Moving them into the real world takes patience, cleanliness, and a slow transition.

Phase 1: Preparation (Before the Box Arrives)

Do not wait until your plants arrive. Having a clean setup ready greatly improves survival rates.

• Substrate: Fluval Stratum (highly recommended for Alocasia) or high-quality peat plugs
• Containers: Small clear 2-inch cups with drainage holes or a seedling tray
• Humidity Dome: Clear plastic bin, prop box, or humidity chamber
• Sterilization: 70% isopropyl alcohol and distilled water
• Fungicide (optional): Mild solution such as Physan 20 or diluted cinnamon water

Phase 2: Unboxing & Cleaning (Day 1)

The goal is to remove all agar (the jelly-like growth medium). Any agar left on the roots can grow mold within hours once exposed to air.

• Gentle removal: Carefully remove plants using clean hands or tweezers
• Lukewarm bath: Swish roots in lukewarm distilled water to loosen agar
• Squeaky clean rule: Use a soft paintbrush to remove every trace of jelly
• Disinfect: Dip for 30 seconds in diluted fungicide or 1:10 hydrogen peroxide solution
• Dry: Pat roots dry gently with a clean paper towel

Phase 3: Potting & Substrate

Alocasias like moisture but need oxygen at the roots. Think “wet feet, dry ankles.”

• Fill the container with Fluval Stratum
• Bury the roots while keeping the crown slightly above the surface
• Use distilled water only
• Keep the substrate moist, not flooded

Phase 4: Humidity Dome (Weeks 1–4)

This is the most critical stage. TC plants do not yet have a protective leaf coating and lose moisture very quickly.

• Seal: Place plants in a clear bin and mist the inside (not the leaves)
• Light: Bright, indirect light or 12–14 hours under a grow light
• Avoid sun: Direct sunlight can overheat plants inside a dome

Humidity Timeline

• Week 1: Lid fully closed at all times
• Week 2: Open lid for 5 minutes daily
• Week 3: Crack lid slightly to reduce humidity to ~80%
• Week 4: Gradually increase airflow until ambient humidity is reached

Expert Tips for Alocasia TC

• No fertilizer at first: Wait until a new leaf forms, then use ¼ strength fertilizer
• Temperature: Keep warm at 70–80°F (21–27°C)
• Leaf melt: Remove mushy leaves immediately with sterilized scissors
• Normal loss: The oldest leaf may die — this is normal if the corm stays firm
